Q2 2026 Earnings Call Summary

Q2 2026
Earnings Call Date:Oct 30, 2026|
% Change Since:

Earnings Call Sentiment|Positive
The call was strongly positive overall. Revenue and core operating profit grew substantially, ENHERTU and DATROWAY showed strong commercial momentum, and the company reported multiple regulatory, clinical and strategic achievements. The main negative items were the withdrawn I-DXd accelerated approval filing, the delayed AVANZAR readout, regional revenue declines, higher expenses, CMO-related payments and the previously disclosed financial reporting corrections. These challenges were material but outweighed by the breadth and scale of the company's operating and pipeline progress.
Company Guidance
Daiichi Sankyo said its consolidated forecast for FY 2026 remains unchanged from the forecast announced in July, with progress on track toward achieving record high full year revenue and a significant year-on-year increase in full year operating profit; JPY 80 billion of CMO compensation fee related to calendar year 2028 is expected to be booked in the third quarter or later this fiscal year, with the total amount projected remaining the same, and the company said this is “more heavy as a matter of probability in the fourth quarter,” while ENHERTU sales milestone of more than JPY 90 billion is expected to be booked in the fourth quarter. The company expects “continued momentum throughout the second half of this fiscal year” for DATROWAY, expects third-quarter profits to be bigger and fourth-quarter profits weaker, with expenses expected to increase from the third quarter to the fourth quarter, and plans to deliver JPY 200 billion cost optimization over 5 years, with targeted benefits exceeding JPY 200 billion over 5 years. AVANZAR is expected to report in the first quarter of calendar year 2027, TROPION-Lung07 in the first half calendar year of 2027, more than 20 registrational trials are expected to be read out over the next several years, and one global commercialization organization will operate effective April 2027.
Revenue Surpassed JPY 1 Trillion
FY 2026 second-quarter revenue increased by JPY 191.9 billion, or 19.7% year-over-year, to JPY 1,167.2 billion. Management said strong growth in ENHERTU and DATROWAY drove revenue past the JPY 1 trillion mark by the second quarter and that the company remains on track toward record-high full-year revenue.
Core Operating Profit Growth
Core operating profit increased by JPY 70.6 billion, or 46.9% year-over-year, to JPY 221.3 billion. Operating profit, including noncore income and expenses, increased by JPY 49.6 billion, or 34.4%, to JPY 193.8 billion.
Full-Year Forecast Maintained
The consolidated FY 2026 forecast remains unchanged from the forecast announced in July. Management stated that the company is well on track toward a significant year-on-year increase in full-year operating profit.
ENHERTU Delivered Broad-Based Global Growth
Global ENHERTU product sales reached JPY 464.5 billion, increasing by JPY 146.1 billion, or 45.9% year-over-year. Growth was 55% in the U.S., 49% in ASCA, 37% in Japan and 26% in Europe, supported by deeper penetration of existing indications, expansion into new indications and broader patient access.
ENHERTU Expanded Into Earlier-Line Treatment
ENHERTU was launched in the United States in both neoadjuvant and adjuvant early-stage breast cancer settings based on DESTINY-Breast11 and DESTINY-Breast05. Management said adoption is increasing month after month in both settings and expects ENHERTU to become the market share leader and new standard of care in those settings.
Additional ENHERTU Approvals and Reimbursement
DESTINY-Breast09 led to ENHERTU approvals in China in August and Japan and the EU in September. DESTINY-Breast05 led to approvals in China and the EU in October. ENHERTU also received approval in China for HER2 pan-tumor solid tumors, bringing the product to 10 indications in China. In the United Kingdom, ENHERTU achieved public reimbursement for HER2-low metastatic breast cancer after chemotherapy, providing access to approximately 1,000 patients.
DATROWAY Sales Accelerated
DATROWAY global product sales reached JPY 48.3 billion, an increase of JPY 32.6 billion year-over-year. Management said growth accelerated sequentially during the quarter and expects continued momentum throughout the second half of the fiscal year.
DATROWAY Clinical and Geographic Expansion
DATROWAY market share continued to expand in U.S. EGFR-mutated non-small cell lung cancer. Its first-line metastatic triple-negative breast cancer launch is progressing well, with market penetration and new patient starts increasing every month. DATROWAY was approved in Japan on September 16 for first-line metastatic triple-negative breast cancer, achieved public reimbursement in Germany, and launched in Brazil in August.
Positive DATROWAY CNS Data
In TROPION-Breast02, DATROWAY significantly improved progression-free survival, response rates and CNS disease control versus investigator's choice of chemotherapy in triple-negative breast cancer patients with baseline brain metastases. Median CNS progression-free survival was 9.5 months with DATROWAY versus 2.6 months with chemotherapy.
Strategic Summit Collaboration
Daiichi Sankyo and AstraZeneca entered a collaboration with Summit Therapeutics to evaluate DATROWAY in combination with ivonescimab across multiple tumor types, including lung and breast cancers. The collaboration will begin with a Phase III study in first-line triple-negative breast cancer.
DESTINY-Lung04 Demonstrated Progression-Free Survival Benefit
In first-line advanced HER2-mutated non-small cell lung cancer, ENHERTU demonstrated a statistically significant progression-free survival improvement versus platinum pemetrexed doublet plus pembrolizumab, with a hazard ratio of 0.63. Median progression-free survival was 14.3 months versus 8.3 months, representing a 6-month improvement and a 37% reduction in the risk of disease progression.
Further R&D and Regulatory Progress
DATROWAY received approvals in the U.S. in May 2026, the EU in July, Japan in September and China in October for first-line metastatic or unresectable triple-negative breast cancer. TROPION-Urothelial04 began with its first subject dosed in August. The company also continued to advance multiple ovarian cancer studies and reported that REJOICE-Ovarian01 data showed a confirmed overall response rate of 49.7%, median progression-free survival of 7.9 months and low-grade ILDs of 5.8%, with no grade 5 ILDs at the 5.6 milligrams per kilogram dose.
Operational Excellence Program Progress
The company is advancing external expense optimization, AI-enabled process transformation and structural transformation. Management said it is developing detailed plans for high-impact profitability initiatives and is targeting JPY 200 billion of cost optimization, with internally targeted benefits exceeding JPY 200 billion over five years.
Continued Investment in R&D and Future Growth
R&D expenses increased by JPY 23.2 billion, reflecting the weaker yen and increased investment associated with development progress for five DXd ADCs and NextWave R&D. Management also said subsequent-generation ADC programs involving linker-payload relationships and different cytotoxic payloads are moving forward and are close to the clinic.
Global Commercialization Reorganization Planned
Effective April 2027, Daiichi Sankyo plans to operate as one global commercialization organization, bringing regional businesses and global commercial functions together under unified accountability. Management said the change is intended to support faster decisions, more efficient resource allocation, stronger launch readiness and more consistent execution across markets.
Recognition for ENHERTU
ENHERTU received the International Prix Galien Award for best technology product, following the National Prix Galien USA Award for Best Technology Product in 2024.
AVANZAR Futility Threshold Passed
Management stated that no interim PFS analysis had been conducted for AVANZAR and that the study had passed futility, with no concerns about futility in the specific patient population.
